An Arahidopsis mutant dwf4 was found to be defective in the 22hydroxylation step (Choe et al, 1997). The mutant plants could be rescued by application of 22-hydroxylated BS including cathasterone and 6- deoxocathasterone. The gene DWF4 responsible for the mutation was cloned. The corresponding enzyme (designated CYP90B1) was shown to be 43% identical to the enzyme CYP90A involved in the 23-hydroxylation of steroids. Further studies have led to the identification of cbbl, cbbS (Kauschmann et al, 1996a,b, 1997), and dim (Klahre and Chua, 1996; Klahre et aL, 1997) BS deficient mutants.
In Table VI data of fragmentation patterns of the major natural BS are given. The disadvantage of 70 eV EI mass spectra is the absence or the extremely low intensity of the molecular ion peak. Other ionization methods such as chemical ionization (CI), field desorption (FD), or fast atom bombardment (FAB) can be applied to overcome this problem. Most characteristic in CI mass spectra are the molecular ion peaks [M+H]^. Dehydration peaks corresponding to the loss of one or two molecules of water are present also.
